研究伞枝犁头霉D1利用蔗糖生产α-酮戊二酸的发酵条件优化。结果表明:1)最佳发酵培养基组成为:蔗糖40 g/L,NaNO3 1 g/L,KH2PO4 1.36 g/L,MgSO4·7H2O 0.2 g/L,FeSO4·7H2O 0.01 g/L,NaCl 2 g/L,玉米浆0.5 g/L,CaCO3 40 g/L,叶酸 2 g/L。2)最佳培养条件为:250 mL摇瓶装液25%,培养温度37℃和转速200 r/min。在此条件下培养84 h,伞枝犁头霉产α-酮戊二酸量最大达14.0 g/L,产酸速率为0.17 g/(L·h),对蔗糖的转化率为34.9%。伞枝犁头霉D1能够有效利用蔗糖发酵产α-酮戊二酸,具有较好的科研价值和工业应用前景。 |

Optimization of fermentation conditions of α-ketoglutarate production from sucrose by Absidia corymbifera |

The present work studied the optimization of α-ketoglutarate production from sucrose by Absidia corymbifera(Lichtheimia corymbifera) D1.Based on single factor experiments,the optimum fermentation medium was found to be 40 g/L sucrose,1 g/L NaNO3,1.36 g/L KH2PO4,0.2 g/L MgSO4·7H2O,0.01 g/L FeSO4·7H2O,2 g/L NaCl,0.5 g/L corn syrup and 40 g/L CaCO3;and the optimal incubation conditions were:25% loading volume of 250 mL flask at 37℃,200 r/min for 84 h.Under the optimal fermentation conditions,the production of α-ketoglutaric acid from sucrose was up to 14.0 g/L,with a conversion of 34.9%.The volumetric production rate was up to 0.17 g/(L·h).Hence,Absidia corymbifera D1 was efficient in utilizing sucrose for α-ketoglutarate production,which shows scientific value and a good prospect for industrial applications. |
