为深入研究绒山羊RORα基因的结构及功能,利用cDNA末端快速扩增法(RACE)结合转录组获得了绒山羊(Capra hircus)维甲酸相关孤核受体(retinoic acid receptor-related orphan receptorα,RORα)基因的2个亚型:RORα1和RORα2(GenBank登录号分别是HM_061155 和 HM_358053)。序列分析表明,RORα1序列全长1 620 bp,包括5'端非翻译区(untranslated region,UTR) 29 bp、3'端非翻译区211 bp和开放阅读框(open reading frame,ORF) 1 380 bp,共编码459个氨基酸,分子质量约为52.3 ku,理论等电点6.25。RORα2序列全长1 694 bp,包括5'UTR 76 bp、3'UTR 211 bp和ORF 1 407 bp,共编码468个氨基酸;与人RORα4的长度相同,相似性达99%,分子质量约为53.4 ku,理论等电点为6.37。结果显示:1) RORα1氨基酸序列缺少大多数核受体具有的调节区(A/B区),预测的二级结构少一个β-折叠基序(YFV);2) RORα的DNA结合区到配体结合区部分属于纯化选择,A/B区在长度和序列方面变异都很大。 |

There are three main receptors in the skin for melatonin,MT1,MT2 and RORα while only the RORα was expressed in goat.In order to study the function and structure of RORα in Cashmere goat,we combined rapid amplification of cDNA ends (RACE) and transcriptome to obtain two isoforms of RORα gene,as accession No.HM_061155 and HM_358053.Sequence analysis showed that the length of the RORα1 cDNA sequence was 1 620 bp,including 5'UTR (Untranslated Region) 29 bp,3' untranslated region of 211 bp and an open reading frame (open reading frame,ORF) 1 380 bp,encoding 459 amino acids,molecular weight of about 52.3 ku,theoretical isoelectric point of 6.25,and the length of the RORα2 cDNA sequence was 1 694 bp,including 5'UTR 76 bp,3'UTR of 211 bp and an ORF 1 407 bp,encoding 468 amino acids is similar to the isoform D of RORα in human with 99% sequence identity,molecular weight of about 53.4 ku,theoretical isoelectric point of 6.25.The goat RORα-LBD tertiary structure was predicted to be similar as the structure of human RORα-LBD.RORα1 lacks A/B region which contains the motif of strands (YFV) compared with RORα2,and result in different third structure.Phylogenetic analysis showed that the region from DBD to LBD of RORα is under purifying selection,and the A/B region is high-diversity for sequence and length.The expression of RORα in different tissues could be regulated by changing the length and structure of the A/B region. |
