采用RT-PCR和RLM-RACE方法从单子叶盐生植物獐茅(Aeluropus littoralis (Gouan) Parl)中克隆了质膜Na+/H+逆向转运蛋白基因(AlSOS1,JN936862),并对其基因序列进行了分析。Southern杂交试验显示:AlSOS1基因在獐茅基因组中以单拷贝形式存在。AlSOS1基因cDNA全长3 641 bp,其中编码区3 420 bp,编码一个1 139氨基酸的蛋白。AlSOS1编码蛋白与芦苇、水稻质膜Na+/H+逆向转运蛋白亲缘关系最为接近,达到82%和77%,与双子叶植物拟南芥的亲缘关系仅为58%;系统发育分析显示AlSOS1基因编码蛋白与其他植物质膜型Na+/H+逆向转运蛋白属于同一分支,而与液泡型Na+/H+逆向转运蛋白属于不同的分支;疏水性分析显示AlSOS1基因编码蛋白N-末端具有12个跨膜结构域,C-末端具有一个很长且面向质膜内腔的亲水尾部。这是獐茅SOS1基因编码区首次被完整克隆,将进一步应用于单子叶农作物耐盐性状改良的基因工程之中。 |

Abstract: |
A plasma membrane Na+/H+ antiporter gene (AlSOS1,JN936862) was isolated from Aeluropus littoralis (Gouan) Parl,a graminaceous halophyte.Southern blot analysis indicated that there was only one copy AlSOS1 gene in the A.littoralis genome.The AlSOS1 sequence contained 3 641 bp with an open read frame of 3 420 bp and the deduced transcripts encoding 1 139 amino acids shared a relative high homology with that of reed(82%) and rice(77%),while a low homology with that of dicot plants as Arabidopsis(58%).A phylogenetic analysis of a number of Na+/H+ antiporter was shown AlSOS1 formed a clade with the most closely related plant SOS1 homolog,which was distinct from the cluster of vacuolar Na+/H+ antiporters.Through hydrophobicity plot analysis,the AlSOS1 protein was predicted containing 12 transmembrane domains in the N-terminal part and a long hydrophilic cytoplasmic tail in the C-terminal part.In a word,the AlSOS1 cDNA was first cloned completely,and suggested the potential utilization for manipulating monocot crops for cultivation under high-salt conditions. |
