从超高压灭菌的瓜汁中筛选出 1株产高分子聚合物的芽孢杆菌B53 。经紫外扫描、高效液相色谱分析及SDS PAGE分子量表征 ,确定该高分子是由谷氨酸单一成分组成的聚合物 ,最大吸收波长为 2 12nm ,分子质量集中在 5 70~ 6 6 9ku ,呈多分子质量聚集体形式 ,不具备典型肽或蛋白质特征。依据B53 的菌株形态、生理生化特征、G +C含量 (摩尔分数 )及 16SrRNA基因序列分析结果 ,将芽孢杆菌B53 菌株鉴定为枯草芽孢杆菌。 |

Isolation and identification of a Bacillus sp. producing poly(γ-glutamic acid) |

Abstract: |
A Bacillus strain isolated from cantaloupe juice treated by superhigh-pressure sterile was shown to produce a high molecular polymer. The purified polymer, was a homopolymer of glutamic acid and had an absorption peak at 212nm. The molecular mass was 570-669ku detected by SDS-PAGE electrophoresis and had no typical structure of protein. With the following identifications of colony morphology, physiological and biochemistry experiments, G+C content and 16S rRNA gene sequence as well, the strain was identified as B. subtilis. |
