以甘薯7个品种及野生种 Ipomoea leucantha(2x)的花药为外植体,在含有2,4-D2mg/L+KT2mg/L+IAA2mg/L 的 MS 培养基上,有6个品种及 I.leucantha 诱导出愈伤组织,而且频率较高。甘薯花药愈伤组织诱导率在基因型之间有差异,向阳黄品种出愈率在7个甘薯品种中最高达9.7%,而甘薯近缘二倍体野生种 I.leucantha 愈伤组织诱导频率高于六倍体甘薯,达11.9%。形成的花药愈伤组织需立即转移到愈伤组织增殖培养基中。在分化培养基上,甘薯两个品种和野生种的花药愈伤组织分化出根。I.leucantha 愈伤组织根的分化率高于甘薯品种,达18.8%。 |

Anther as the explant from 7 varieties of sweet potato and its relative sp.I. leucantha(2x)were cultured on MS basic medium supplemented with 2,4-D 2mg/L+KT2 mg/L+IAA 2mg/L.Except one variety,calli were formed from all explants.But the rate of callus induction was different among different genotypes,that of Xiang Yang Huang was 9.7%.while that of I.leucantha reached 11.9%.Anther callus must be transferred to multipli- cation medium.Root development was obtained when calli were cultured on 1/2 MS medium supp... |
